Mehr als zehn Jahre nach den Ereignissen des ersten Films haben Jake (Sam Worthington) und Neytiri (Zoe Saldana) eine Familie gegründet. Ihre Kinder sind Neteyam (Jamie Flatters), Lo'ak (Britain Dalton) und Tuktirey (Trinity Bliss), der adoptierte Menschenjunge Miles "Spider" Socorro (Jack Champion) und die adoptierte Na'vi-Teenagerin Kiri (Sigourney Weaver). Doch ihre Heimat ist weiterhin nicht sicher vor den Machenschaften der Resources Development Administration (RDA), die fremde Planeten ausbeutet und dort für die Menschen wichtige Ressourcen abbaut. Nach dem Tod von Colonel Miles Quaritch (Stephen Lang) hat nun General Ardmore (Edie Falco) die Kontrolle über die RDA übernommen. Bald sehen sich Neytiri, Jake und ihre Familie gezwungen, ihr Zuhause zu verlassen und beim Na'vi-Stamm der Metkayina Zuflucht zu suchen, der an den Küsten und Meeren des Mondes Pandora lebt und von der schwangeren Ronal (Kate Winslet) und ihrem Mann Tonowari (Cliff Curtis) angeführt wird. Im Sequel zu „Avatar“ entspricht der zeitliche Abstand, der zwischen Teil 1 und Teil 2 liegt, auch ungefähr dem Zeitsprung in der Handlung zwischen beiden Filmen.
